Postdoctoral fellow in The role of STK25 in regulation of metabolic profile of the body Type of employment: Fixed-term employment, 24 months Extent: 100 % Location: Institute of Medicine, Gothenburg First day of employment: 2016-09-15 Reference number: PER 2016/333 Our primary research focus at Department of Molecular and Clinical Medicine is on metabolism, diabetes, obesity, and cardiovascular disease. We support and promote research and teaching in translational medicine to improve health and to benefit society. The focus of Dr. Margit Mahlapuu’s research over the last decade has been to characterize the role of metabolic master regulator – AMP-activated protein kinase (AMPK) - in control of energy homeostasis. A gene array study showed that in AMPK?3-/- mice, the expression of serine/threonine kinase 25 (STK25) was significantly altered, which identified STK25 as a novel target for AMPK and a possible regulator of metabolism. The main focus of the research group now is to characterize the role of STK25 in regulation of whole body metabolism, combining in vitro assays with studies in animal models and in unique patient cohorts. Subject area The role of STK25 in regulation of metabolic profile of the body Specific subject description Type 2 diabetes (T2D) is characterized by hyperinsulinaemia, hyperglycaemia and dyslipidaemia. With its steep increase in incidence, T2D is becoming one of the major global threats to human health. In industrialized countries, the total burden of diabetes and related complications is approximately 10% of the total health care costs. Since the efficacy and patient compliance of existing treatments are in many cases unsatisfactory, there is a pronounced need for novel targets, which could complement current treatment. Our recent studies suggest that STK25 represents a novel key regulator of glucose and lipid metabolism in peripheral tissues prone to diabetic damage. We have found that STK25 is localized on the surface of hepatic lipid droplets, and it controls the metabolic balance of lipid storage versus lipid utilization and thereby regulates the ectopic lipid deposition in the liver. We have now initiated the detailed studies investigating the role of STK25 in other metabolic organs such as adipose tissue, skeletal muscle and pancreas. Job assignments The applicant will be investigating the role of STK25 based on both in vitro and in vivo experiments, involving unique mouse models, human cells as well as patient material. Eligibility To qualify as a postdoctoral researcher the applicant should hold a PhD in a relevant subject. Since postdoctoral constitutes a qualifying appointment for junior researchers, we aim to target those who have a doctoral degree not older than 3 years from the application deadline. The applicant must also have a vast experience with in vivo studies in mice, and be able to perform different metabolic studies such as glucose and insulin tolerance tests, euglycemic hyperinsulinemic clamps, lipid secretion and fatty acid uptake. In addition, experience in cell culture and ex vivo assays is considered an advantage.
