Reference number: UR 2016/438. Project title: Preventing chronic neurotoxicity in childhood cancer survivors. Job assignments: Brain tumours are one of the most common types of paediatric cancer. Today, thanks to improved treatments, approximately 80 % of the patients survive, but the treatment itself can cause lifelong adverse effects. Radiotherapy is one of the curative methods used. Irradiation is effective in treating cancer but can also damage the healthy brain tissue permanently which can cause impaired cognitive function. The paediatric brain undergoes a series of developmental stages during its maturation, which makes young age a risk factor for radiation-induced damage. Another important factor for late effects is believed to be a chronic alteration of the chemical milieu which makes it harder for the brain to heal. We have seen that the levels of APP (amyloid precursor protein) in the CSF was progressively reduced in patients that had received prophylactic cranial radiotherapy. This indicates that APP may play a role in the late effects after irradiation. Further studies in mice show that APP is more abundant in the thalamus after cranial irradiation. The purpose of this PhD project will be to further elucidate APP's role in the juvenile brain's response after irradiation. The project will also include evaluation of potentially alleviating effects of pharmaceutical interventions after irradiation. This will primarily be achieved through different molecular methods such as qRT-PCR, Western Blot, ELISA and immunohistochemistry. Our research group has ongoing preclinical and clinical projects and consists of pharmacologists, physicians and molecular biologists. We are now looking for a dedicated PhD student who is methodical, conscientious, creative, can take their own initiatives and has a genuine interest in science. Laboratory knowledge, experience in different molecular techniques and practical experience from working with laboratory animals are creditable. The PhD student must be able to communicate well in English. Personal suitability will be of great importance. To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
